The Giants are looking for their second win of the season, just like when they visited the then-winless 49ers last season and lost to quarterback C.J. Beathard. Mullens is even more of an unknown now -- entering his second career start -- than Beathard was then.
Mullens threw three touchdown passes in a win against the Raiders last week, keeping the job over Beathard.
"It gets harder and harder. It doesn't get easier," 49ers coach Kyle Shanahan said. "It will be a big challenge this week versus the Giants. They do a hell of a job defensively with their scheme and with their players."
Meanwhile, it's gotten very hard for Manning, a two-time Super Bowl MVP trying to hold onto his starting job as the Giants have lost 20 of their last 24 games. What's missing?
"I think it's just scoring more points in the red zone," Manning said. "I feel like we're moving the ball well. I feel like we've made improvements between the first four games, and the second four games. Getting yards and moving the ball, and getting down there, and being a little bit more explosive. Now, we need to translate that into more touchdowns."
